I think promo is short for promotion. Many manga "promos" is made to promote the novel, which means that the manga won't be continued and only one chapter will ever be released. So when someone says "It's not a promo!" they're basically saying that the manga will be continued and not just stop at being a promotion of something else :)
Promo means that it’s usually only 1 chapter to promote the novel. If it’s not promo that means the novel story is already adopted by an artist and there will be the webcomic version that will be an ongoing series
