The condition was that the Esper's paired guide should be killed.
In one of the chapters (I think it's chapter 62 or 63), there is a story about a guide who was an illegitimate son of a wealthy family and was forced to sleep with any powerful esper (for guiding) as a way to increase the family's power. He then met a doctor who was an esper and obeyed everything the guide told him like a dog. It can be the doctor's backstory, but it is not known yet. Maybe this is the reason why even though he hates espers he wants to become one.
It can also be the reason why he wanted to be an esper but couldn't and created Lee Hansol instead.
Ji Gwanyeong is an SSS class esper who has a match rate of 151.20% which is unprecedented. The doctor wanted to take this chance and study his bond with Taehun to create a perfect esper and maybe turn himself into one.
The center has data of all the registered guides, espers, and their pairing. He used the connection between the center and the guides to create a sound wave that lets him manipulate the guides' thoughts and actions. (Our brains are a huge mesh of brainwaves and sine waves that can actually be manipulated - it's the theory behind hypnosis and hypnotherapy (●'◡'●)ノ ) That's how he killed so many guides and could threaten the center with mass murder.

MC is the ultimate guide since he is a bonded guide to one of the strongest espers in the world if not the strongest. Doc can’t kill him because he’s a vital part of the research. Both MC and ML are vital to the docs plans. The doc probably doesn’t have much time left considering he’s now an esper that’s about to rampage from not having good enough guiding for god knows how long he’s been an esper.

Because that guide can only work on bonded espers. He can steal a guiding compatibility rating but that’s it. He’s not actually an outstanding guide. That’s why MC is a far better ideal guide.

As an esper, he can no longer guide and therefore he cannot self regulate. He hates espers because of how reliant they are on guides who he views as beneath him because they aren’t “powerful” like espers.
The reason he ended up turning hansol into guide is because he was trying to make a super esper and failed and decided to pivot to make a super guide aka one that could guide him and give him more time to figure out how to make a super esper. Also from a scientific perspective, it’s actually better to start off from a stable base than an unstable one. I won’t bore you with the science but if the magic number is 8 and a guide is 7/8 stable and an esper is 1/8 stable, it’s easier to start from 7 and work up to 8 than start at 1 and work up to 8. That’s how atomic and molecular science works. You always want to reach 8 if you want stability.

I agree with the scientific example. It's better to accept one electron to reach stability rather than gain 7 electrons to try and reach stability in the that shell.
