The Leader knows what Ulysses intentions inside that place. As we all know Ulysses is fond of experimenting chemicals like drugs/poisons/medicine and etc since he is originally a son of the doctors who died at the first season. Ulysses intentionally go there at that place to steal all the drugs experimented by the people there so he can experiment it on his own and claim it as his.

Yes. She meant it like she already knows about the drugs and the place (kinda observing and planning how to raid it) But as for Ulysses who got kidnapped, I'm not really sure. I think the Leader just let go of Ulysses coz she knows Ulysses doesn't live there.
