I think the prince do know that "Madame Chen" is a he, but since manhwas do not follow exactly the flow of the novels it was just implied and was never shown how the prince knew since whenever the prince addresses to the public about Chen, the dialogue box uses "she" with the quotation to imply that the Prince DOES know.

yes. the Prince know. And besides, it really is impossible for the Prince not to know if they had already, you know, slept almost every night as soon as Chen got in the palace. It was just Chen who did not know the prince is already aware that he is a guy ╮(￣▽￣)╭
