Guide-Verse is basically a world setting where one day Portals/Dungeons just randomly start opening everywhere in the world.
Espers are humans with all kinds of superpowers that go into these portals and defeat the monsters inside. If they don't do this monsters will start invading earth through the portal.
Some dungeons/portals don't close/disappear if the monsters are not defeated.
The other humans that have special powers are called Guides. They basically help Espers control their supernatural powers because if they don't do this it will harm the Esper.
Espers can go without Guiding with the help of medication, but if they keep using the Esper power it won't be enough at some point.
Both Espers and Guides need each other. Espers need Guides to stay healthy and Guides need Espers because of their ability to defeat monsters.
Both Espers and Guides can be categorized between E-SS, with E being the weakest and SS being the strongest.
