The first is a more sarcastic tone, like “you know you had no reason to roofie me, right? That was a dick move.” which is how I assume a lot of people read it as. The second is the more literal meaning “I know and believe that you had no reason to roofie my drink, it’s not something you would do” which is how Uno intended to say it.
