It's more the reaction that Haruki has to her coming home. He freaks out that she came home early, refers to Shirayuki as a just a classmate and not a friend as if to show his sister that he keeps him at a distance and they don't have a friendly relationship. She also immediately went to spy on Haruki and Shirayuki and has a past of following him around as a child, which can be innocent, but in tandem with the other things it's added suspicion. It points less to her being explicitly mean to him and more that she's also possibly obsessive about her brother and has maybe a past of turning his friends away.
