I have two hypothesis... 1. Sis knew everything and left after having the twins, purposely planting the hint that lead Ryogo to going to her brother. 2. She couldn’t stand Ryogo’s workaholic nature and was too much of a free spirit to be tied down, so she up and left.

Personally I think it’s hypothesis 1.
